The Malvern Zetasizer-nano is a multi purpose analyser combining
simplicity and precision. This desktop system combines the capability
of static and dynamic light scattering (SLS and DLS) plus Zeta potential.
Novel optics provides exceptional sensitivity to size molecules
smaller than 1000 Dalton's. Sizing of nano-particles from 0.6 to
6000nm is now extremely simple and rapid.
Rapid Crystal screening
The Zetasizer-nano can dramatically reduce the screening time needed
to optimise protein crystal growth. SLS can be used to determine
the second viral coefficient (A2), a property that describes the
strength of protein-solvent interaction and correlates with solubility.
The determination of A2 for various protein solutions can help predict
which is the most suited to facilitate crystal growth. DLS is complimentary
since it is ideal for distinguishing which solutions are unlikely
to yield crystals due to impurities or non-specific aggregates.
Easy molecular weight
Since the majority of bio-molecules and polymers are less than
100nm, molecular weight can be determined by DLS. If absolute molecular
weight is required measurements are made at a number of concentrations.
However for fast and easy screening between monomers and dimers
only a single measurement is needed. The technique is very sensitive
to small changes in protein size, enabling detection of small conformational
changes.

Dual parameter melting points
Trend measurements of size or zeta potential as a function of temperature
allow for the determination of protein melting points and can give
information on heat stability and conformational change. The temperature
range is 2 to 90 degrees C.
Zeta potential
Zeta potential is measured by the patented M3-PALS technology,
giving the highest ever sensitivity and resolution. Cross contamination
is eliminated by means of a unique disposable capillary cell.
Chromatography detector
A new chromatography flow-mode for the Zetasizer-nano enables
it to be also used as a highly sensitive size exclusion chromatography
detector. This enhanced capability provides a major new tool
for protein and other biomolecular research.

chromatography flow-mode combines dynamic light scattering
with size exclusion chromatography in order to perform Absolute
Size Exclusion Chromatography. This technique allows protein size
to be measured directly without the need for column calibration.
Molecular weight standards are no longer required to identify oligomeric
species or to determine protein purity.
The flow cell in the Zetasizer is connected after the last detector
in a size exclusion chromatography system. The software makes the
entire measurement process simple and straightforward. For the majority
of protein separations no extra parameters are required. Size is
plotted in real time as material is eluted from the column. The
molecular weight of each peak is calculated automatically at the
end of the measurement.
